藉由增加促進beta-cell分泌胰島素的腸泌素或是抑制腸泌素降解而增加腸泌素而增加胰島素。GLP-1 (Glucagon-Like Peptide-1)(此類的藥物較常見)、GIP (Glucose-dependent Insulinotropic Polypeptide 或 Gastric Inhibitory Polypeptide)。增加促進beta-cell分泌胰島素的腸泌素可能有增加胰臟癌機率的疑慮；抑制腸泌素降解的藥品有增加甲狀腺C細胞癌的疑慮。目前學界認為有恢復beta-cell的功能而根治糖尿病的可能性。有胰臟癌前病變的人和急性胰臟炎患者不能用。
